Star Activity

Many restriction enzymes have been reported to cleave sequences that are
similar to their defined recognition site.  This is called star activity
and is usually found when restriction enzymes are used under non-standard
conditions.  Star activity was first reported for EcoRI, when it is
incubated in low ionic strength, high pH buffer.  Other conditions that
can induce star activity include substituting Mn2+ for Mg2+, high levels
of organic solvents such as glycerol, DMSO or some detergents.  Not all
restriction enzymes have been tested for star activity, and only a few
have been examined systematically.

Those for which star activity has been seen experimentally are listed
below, with references.  It should be noted that typically the sites
recognized under star conditions differ from the canonical recognition
sequence at one position.  Thus under star conditions, EcoRI (canonical
recognition sequence: GAATTC) will probably cleave NAATTC, GNATTC, GANTTC,
GAANTC, GAATNC and GAATTN.  Note that not all possibilities have been
tested experimentally either in this case or in any of the other cases
listed below.

As a general guide, star activity is likely to result in cleavage at
sequences that differ by one base from the canonical recognition sequence.
However, one can expect different star sites to be cleaved at different
rates.  It is probable that under sufficiently extreme buffer conditions,
most, if not all, restriction enzymes can be persuaded to cleave at
non-canonical sites and hence would show star activity.
